Characteristics of the participants in discovery and replication analyses

A flowchart of the primary analyses is presented in Fig. 1. In the discovery analysis, the total sample size was 7277 from three cohorts. The mean age was 46 in the Framingham Heart Study Third Generation cohort (FHS Gen3) and 75 in the Atherosclerosis Risk in Communities (ARIC) study and Cardiovascular Heart Study (CHS). FHS included White participants only. ARIC and CHS included Black and White participants (17% and 14% Black, respectively, Supplementary Data 1). The protein measures were quantified using an aptamer-based platform (SomaScan, Supplementary Data 2).
