Non-invasive in vivo hyperspectral imaging of the retina for potential biomarker use in Alzheimer’s disease

Study design

The primary objective of this study was to determine whether retinal HS imaging could be used to distinguish Aβ PET+ (case) from PET− (control) participants. Accordingly, participants who had undergone brain Aβ PET imaging were recruited for HS imaging of the retina. A spectral analysis method was used to minimise spectral variability and identify a difference between Aβ PET+ cases and PET− controls. The method was further validated against an independent cohort to determine whether the same discrimination performance could be obtained. To understand the underlying components of the main spectral difference between groups, modelling was performed using a combination of spectra of ocular structures that are known to affect ocular reflectance as well as the spectrum of Aβ in solution derived from spectrophotometry.
